摘要:
目的:建立HPLC同时测定羚羊清肺颗粒中绿原酸、栀子苷、橙皮苷、黄芩苷和甘草酸含量的方法;采用Agilent TCC18色谱柱(4.6mm×150mm,5μm),流动相为乙腈(A)-0.6%磷酸水溶液(B),梯度洗脱程序为(0~20min,4%→10%A;20~24min,10%→8%A;24~25min,8%A→10%A;25~28min,10%A→12%A;28~30min,12%A→23%A;30~40min,23%A→30%A;40~45min,30%A→35%A;45~46min,35%→40%A;46~50min,40%→50%A;50~55min,50%→60%A;),流速为1.0m L·min-1,柱温为30oC,检测波长分别为238nm(栀子苷),250nm(甘草酸),280nm(橙皮苷、黄芩苷),327nm(绿原酸)。结果:绿原酸、栀子苷、橙皮苷、黄芩苷和甘草酸进样量分别在0.0470~2.8217μg(r=0.9996),0.02708~1.6247μg(r=0.9996),0.0077~1.5474μg(r=0.9999),0.0235~1.4100μg(r=0.9999),0.0072~0.4333μg(r=0.9996)范围内线性关系良好;平均加样回收率(n=6)分别为96.93%、98.04%、97.10%、101.15%和95.93%,RSD分别为1.62%、1.15%、0.57%、0.80%和1.71%。结论:本法适用于同时测定羚羊清肺颗粒中绿原酸、栀子苷、橙皮苷、黄芩苷和甘草酸的含量。
